The NS5B protein of HCV, which is a RNA-dependent RNA polymerase, plays a crucial role in the replication of the virus. Previous studies have shown that different lengths of NS5B have different enzymatic activities, but the methods for expressing, purifying, and measuring the activity of these proteins are not well established.Objectives:This study aims to establish a new method for expressing, purifying, and measuring the enzymatic activity of different lengths of NS5B in HCV.Methods:The different lengths of NS5B will be cloned into bacterial expression vectors and expressed in Escherichia coli. A variety of purification methods, including nickel affinity chromatography, ion exchange chromatography, and gel filtration chromatography, will be evaluated to obtain pure and active proteins. The enzymatic activity of each NS5B protein will be measured using a fluorescence-based assay, and the data will be analyzed by non-linear regression analysis.Expected Results:We expect to establish a robust method for expressing, purifying, and measuring the activity of different lengths of NS5B in HCV.
