精品文档---下载后可任意编辑α-硫辛酸对硫酸吲哚酚致脐静脉内皮细胞氧化损伤的影响的开题报告摘要:硫酸吲哚酚(SIP)是一种有效的抗氧化剂,已被广泛应用于保健品和药物中,具有良好的生物活性和安全性。然而,SIP 也存在着一定的毒性和副作用。本文旨在讨论 α-硫辛酸(LA)对 SIP 致脐静脉内皮细胞(HUVEC)氧化损伤的影响。通过 MTT 法评估不同浓度(0.1 mM、0.5 mM、1 mM、2 mM、5 mM)LA 处理 HUVEC 的细胞存活率,结果发现 LA 对 HUVEC 没有明显的细胞毒性,在此基础上选取合适的浓度组合(0.5 mM LA + 100 μM SIP)进行后续实验。通过检测SIP 致 HUVEC 内源性抗氧化酶(SOD)活性、总抗氧化能力(T-AOC)和丙二醛(MDA)含量,结果发现 LA 能显著提高 SIP 致 HUVEC的 SOD 活性和 T-AOC,降低 MDA 含量。同时,LA 还能减轻 SIP 引起的细胞凋亡,提高 HUVEC 的迁移能力。综上结果表明,LA 可以保护SIP 致 HUVEC 免受氧化损伤,具有一定的应用价值和进展前景。关键词:硫酸吲哚酚,α-硫辛酸,氧化损伤,内皮细胞,细胞保护Abstract:Indole-3-propionic acid (SIP) is an effective antioxidant that has been widely used in health products and medicines due to its good biological activity and safety. However, SIP also has certain toxicity and side effects. This study aims to investigate the effect of α-lipoic acid (LA) on SIP-induced oxidative damage in human umbilical vein endothelial cells (HUVEC). The cell survival rate of HUVEC was evaluated using the MTT method at different concentrations (0.1 mM, 0.5 mM, 1 mM, 2 mM, 5 mM) of LA treatment, and the results showed that LA had no significant cytotoxicity on HUVEC. Based on this, a suitable concentration combination (0.5 mM LA + 100 μM SIP) was selected for subsequent experiments. By detecting the endogenous antioxidant enzyme (SOD) activity, total antioxidant capacity (T-AOC) and malondialdehyde (MDA) content induced by SIP in HUVEC, it was found that LA could significantly increase SOD activity and T-AOC in SIP-induced HUVEC, and reduce MDA content. At the same time, LA can also alleviate SIP-induced apoptosis and improve the migration ability of HUVEC.
