精品文档---下载后可任意编辑骨髓间充质干细胞治疗大鼠暴发性肝功能衰竭及其机制的初步讨论的开题报告摘要暴发性肝功能衰竭病因复杂,且治疗方法有限。近年来,骨髓间充质干细胞(BMSCs)被广泛讨论,发现其对于肝损伤的修复具有很强的潜力。本讨论旨在探究BMSCs 在治疗大鼠暴发性肝功能衰竭中的作用及其机制。首先,将 40 只 SD 大鼠随机分成 4 组,包括正常对比组、模型组、BMSCs 处理组和寡核苷酸(siRNA)干扰组。通过乙酰氨基酚和二甲双胍诱导肝损伤,建立大鼠暴发性肝功能衰竭模型。接着,将BMSCs 和 siRNA 注射到大鼠体内,分别观察其对肝功能、肝细胞凋亡、肝细胞周期、炎症反应、氧化应激及肝细胞再生的影响,并进一步探究其作用机制。估计本讨论可为探究 BMSCs 的治疗机制提供新的思路,为暴发性肝功能衰竭的临床治疗提供新的方法。关键词:骨髓间充质干细胞;暴发性肝功能衰竭;肝细胞凋亡;肝细胞周期;炎症反应;氧化应激;肝细胞再生AbstractAcute liver failure (ALF) has a complex etiology and limited treatment options. In recent years, bone marrow mesenchymal stem cells (BMSCs) have been extensively studied and found to have strong potential for repairing liver injury. This study aims to investigate the role and mechanism of BMSCs in the treatment of ALF in rats. Firstly, 40 SD rats were randomly divided into 4 groups, including a normal control group, a model group, a BMSCs treatment group, and an siRNA interference group. ALF rats were induced by acetaminophen and metformin, and then BMSCs and siRNA were injected into rats, respectively. The effects on liver function, liver cell apoptosis, liver cell cycle, inflammatory response, oxidative stress, and liver cell regeneration were observed, and their mechanisms were further explored. It is expected that this study can provide new ideas for exploring the therapeutic mechanism of BMSCs and provide new methods for the clinical treatment of ALF.Keywords:bone marrow mesenchymal stem cells; acute liver failure; liver cell apoptosis; liver cell cycle; inflammatory response; oxidative stress; liver cell regeneration
