大鼠移植肝保护管理论文 【摘要】目的探讨川芎嗪（TMP）对大鼠移植肝的保护作用。方法参照 Delriviere 和 Kamada 等方法建立大鼠肝移植模型，将大鼠随机分为 3 组：A 组（假手术组）、B 组（肝移植对比组）和 C组（TMP 干预组）。采纳全自动生化分析仪检测术后血清肝功能(AST、ALT、LDH)水平；应用免疫组化 SP 法结合病理图像分析仪测定肝组织中 Bcl-2、Bax 的表达情况；HE 染色观察其病理形态学变化。结果（1）C 组 AST、ALT、LDH 明显低于 B 组（P0.05）。（3）A 组无明显病理形态改变，B 组术后肝组织出现明显的病理损害，C 组病理变化明显减轻。结论 TMP 可能通过上调 Bcl-2 或 Bcl-2/Bax 表达而减轻肝移植体的低温保存再灌注损伤，对供肝有保护作用。 【关键词】肝移植；保存再灌注损伤；川芎嗪；Bcl-2；Bax Protectiveeffectoftetramethylpyrazineonlivergraftinratlivertransplantation 【Abstract】ObjectiveToinvestigatetheprotectiveeffectoftetramethylpyrazine(TMP)preconditioningonlivergraftinratlivertransplantation.MethodsPerformingtheoperationoflivertransplantationinmaleSprague-DawleyratsreferredtoDelriviereandKamadawithcuffanastomosingmethods，theratswererandomlydividedintothreegroups：Agroup(n=6，shamoperationgroup)，Bgroup(n=30×2，livertransplantationcontrolgroup)，Cgroup(n=30×2，TMPgroup).Theparametersofliverfunction(AST ， ALTandLDH)weretestedbyauto-biochemicalanalyseringroupBandCafterreperfusion0.5h ， 3h， 6h ， 24h ， 48hrespectivelyviatheplasmasamples ；partofthelivertissuewassampledtodeterminetheexpressionofBcl-2andBax.TheparametersingroupAwereexamed3haftertheoperation.TheexpressionofBcl-2andBaxwasdeterminedbyimmunohistochemistytechniquewithpathologicalimaginganalyser.Also ， partsoflivertissuesampleswereusedtodetectthehistopathologicchanges.Results（1）ThelevelsofAST ， ALTandLDHweresignificantlydecreasedingroupCcomparedwiththatingroupB（P＜0.01）afterreperfusion0.5h，3h，6h，24h，48hrespectively.（2）TheexpressionofBcl-2weresignificantlyincreasedingroupCcomparedwiththatingroupB（P＜0.01）afterreperfusion0.5h，3h，6hrespectively.ThedifferentexpressionofBaxbetwe...
