Long term clinical and experimental studies have indicated that the arsenic trioxide is very effective in the treatment of diseases in China..This paper reviews the current research situation of the arsenic trioxide against malignant tumor, exploring the possibilities for the efficacy and mechanism in treatment of malignant tumor, and provides the reference for developing a new field in arsenic trioxide against malignant tumor. 【Key words】 arsenic trioxide；tumor ;therapy ; apoptosis; calcium channel 三氧化二砷，是传统中药砒霜的有效成分之一，祖国医学很早就对其性质做了描述并用于临床。20 世纪 70 年代后，张亭栋等单用 As2O3 注射液用于急性早幼粒细胞白血病的治疗取得显着疗效，许多学者尝试着将其用于其他恶性肿瘤的治疗。随着讨论的不断深化,证明 As2O3 具有良好的抗肿瘤作用,而且已被广泛应用于白血病、骨髓瘤、乳腺癌、肝癌等肿瘤的治疗，取得了良好的临床效果。大量体内和体外实验讨论证明,三氧化二砷对实体瘤同样有显着治疗效果，本文就现阶段内三氧化二砷的抗肿瘤作用机理的实验讨论及临床运用的进展作一综述。 1 As2O3 诱导肿瘤细胞凋亡的几个途径 激活 caspase 家族 讨论表明 AS2O3 的细胞毒作用有 caspase 依赖和非 caspase 依赖两种途径［1］。caspase-3 在半胱氨酸家族中占重要地位，广泛分布于组织中并参加不同细胞的凋亡［2］。As2O3 通过破坏线粒体跨膜电位， 触发了线粒体凋亡途径，导致了 caspase 3 的活化，促使细胞凋亡［3］。有实验认为，线粒体和微管可能是三氧化二砷的主要作用位点,为三氧化二砷发挥凋亡诱导效应的启动因素，caspase-3 途径的激活可能为三氧化二砷诱导口腔鳞癌细胞凋亡的关键途径之一。 ROS 过度生成 砷剂可使细胞内 ROS 生成增多是由于：MPT 开放，线粒体内氧化呼吸链受...
