ChineseJournalofReparativeandReconstructiveSurgery,January2008,Vol.22,No.1·88·三种组织工程鼓膜的构建邓志宏1田勇胜1王锦玲1邱建华1刘源2赵宇2金岩2【摘要】目的构建3种不同材料的组织工程鼓膜,并对其进行生物学特性检测。方法体重30kg健康雄性小白猪1只,制备脱细胞硬脑膜及脱细胞真皮,并从牛皮中提取液态胶原,将第3代豚鼠鼓膜成纤维细胞1×106个/mL,各0.2mL种于3种材料表面进行培养,倒置相差显微镜下观察成纤维细胞的生长状态。培养1周后,将3种材料分别进行扫描电镜及HE染色观察,测定材料厚度及其张力。结果成纤维细胞在3种材料表面生长良好,呈束状平行排列,其中以胶原与脱细胞真皮表面成纤维细胞生长情况较佳。HE染色及扫描电镜下可见成纤维细胞呈单层于3种材料表面连续生长。胶原膜、脱细胞真皮及脱细胞硬脑膜的平均厚度分别为9.4、10.0及10.4μm。张力强度测定,复合成纤维细胞的胶原组织工程鼓膜张力(1.417±0.030)N/mm2,复合成纤维细胞的脱细胞真皮张力为(24.500±2.040)N/mm2,复合成纤维细胞的脱细胞硬脑膜张力为(53.300±2.600)N/mm2;复合成纤维细胞的脱细胞真皮张力与正常豚鼠鼓膜张力26.700N/mm2最接近。结论脱细胞猪真皮为支架复合成纤维细胞构建的组织工程鼓膜,其张力与厚度均与正常豚鼠鼓膜相接近,是构建组织工程鼓膜的理想材料。【关键词】组织工程鼓膜构建体外培养中图分类号:R322.921Q813.01文献标志码:ARECONSTRUCTIONOFTHREEDIFFERENTKINDSOFTYMPANICMEMBRANEINVITROBYTISSUEEN-GINEERINGTECHNIQUE/DENGZhihong1,TIANYongsheng1,WANGJinling1,QIUJianhua1,LIUYuan2,ZHAOYu2,JINYan2.1DepartmentofOtolaryngology,XijingHospital,theFourthMilitaryMedicalUniversity,Xi’anShaanxi,710032,P.R.China;2DepartmentofPathology,CollegeofStmatology,theFourthMilitaryMedicalUniversity.Correpondingauthor:DENGZhihong,E-mail:dengzh@fmmu.edu.cn【Abstract】ObjectiveToreconstructthreedifferentkindsoftympanicmembraneinvitrobytissueengineeringtechnique,andtoexaminetheirhistologicalstructuresandmechanicalproperties.MethodsTheskinandduraofpig(weight30kg)wereprocessedwithhighsatuatedsalineandenzymestomakeextracellularmatrix.Meanwhile,fibroblasts(1×106/mL,0.2mL)wereseededonthesurfaceofthesetwoscaffoldsandcollagen.Thecompositetissueswereculturedinvitrofor1weekandexaminedinhistologicalstructureandmechanicalproperties.ResultsFibroblastsculturedwerespindle–shapedandcouldgrowandattachtothesescaffoldswithaarrangementofsarciniformandparallel.ThereconstructedtissueofECMandcollagenappearedtointegratewellandhadbetterbio-compatibility.Themeanthicknessofthecollagen,theskinandthedura(allcoveredwithfibroblasts)were9.4,10.0and10.4μmrespectively.Thetensionofthecollagenwas(1.417±0.030)N/mm2,oftheacellulardermalmatrixwas(24.500±2.040)N/mm2(beingclosetothetensionofnormaltympanicmembrane,26.700N/mm2),oftheacellulardurawas(53.300±2.600)N/mm2.ConclusionTheresultssuggestthatthetensionandthethinknessofacel-lulardermalmatrixissimilartothenormaltympanicmembraneofguineapig,itisanidealmaterialfortympanoplasty.【Keywords】TissueengineeredtympanicmembraneReconstructionInvitrocultureFoundationitem:NationalNaturalScienceFoundationofChina(30371527)随着组织工程研究的不断深入,组织工程在真皮、骨、软骨、脂肪及血管等领域的研究取得了很大进展,临床上出现了许多类型的组织工程替代物[1-6]。目前,组织工程鼓膜的研究国内外尚处于起步阶段[7-9]。我们的实验拟通过比较3种不同的组织材料,探讨构建组织工程鼓膜的可能性。基金项目:国家自然科学基金资助项目(30371527)作者单位:1西安第四军医大学西京医院耳鼻咽喉科(西安,710032);2西安第四军医大学口腔医院病理科通讯作者:邓志宏,副教授,硕士导师,研究方向:组织工程,E-mail:d...
