摘 要以越南可口革囊星虫为原材料,采用酸法从体壁中提取胶原蛋白,对所提胶原蛋白的理化特性进行研究。通过紫外全波长扫描(UV)、SDS-PAGE 凝胶电泳、氨基酸分析、粘度和变性温度特性测定、凝胶强度、保湿性对胶原蛋白进行分析。结果显示,可口革囊星虫 ASC 紫外扫描波长在 230nm 处有最大吸收峰,SDS-PAGE 电泳结果显示可口革囊星虫 ASC 含有 β、α1、α2 链,属于 I 型胶原蛋白;可口革囊星虫 ASC 中甘氨酸、谷氨酸和脯氨酸含量较高;可口革囊星虫 ASC 变性温度为 33.6℃,具有较好的热稳定性;凝胶强度为 84.65N;保湿率与甘油相比较低。关键词:可口革囊星虫;酸法;胶原蛋白;理化特性Abstract Using Phascolosoma esculentas in Vietnam as raw materials, the collagen was extracted from body wall by acid method, and the physical and chemical properties of the extracted collagen were studied. Collagen was analyzed by ultraviolet full wavelength scanning (UV), SDS-PAGE gel electrophoresis, amino acid analysis, viscosity and denaturation temperature characteristic determination, gel strength and moisture retention. The results showed that the ultraviolet scanning wavelength of ASC of Phanerochaete vietnamese had the maximum absorption peak at 230nm. SDS-PAGE electrophoresis results showed that ASC of Phanerochaete vietnamese contained β, α1 and α2 chains, belonging to type I collagen. The contents of glycine, glutamic acid and proline in ASC of Phanerochaete vietnamese are relatively high. ASC denaturation temperature of Phanerochaete vietnamese is 33.6, which℃ has good thermal stability. The gel strength was 84.65 N; The moisture retention rate is lower than that of glycerol.
