This study aims to identify the key sequence of GhPAP1 transcription activation region and investigate its effect on anthocyanin regulation. The transcription activation region of GhPAP1 was predicted by bioinformatics methods, and the activity of the region was verified by yeast two-hybrid and firefly luciferase reporter gene systems. Further, the reverse transcription fluorescent quantitative PCR technique was used to detect the expression of different mutants on anthocyanin biosynthesis genes. The final results showed that the GhPAP1 transcription activation region contains two nucleotide fragments GTTA and GTCT, and mutations in these fragments will lead to down-regulation of the expression of anthocyanin biosynthesis genes. The study showed that the GTAA and GTCT nucleotide fragments in the GhPAP1 transcription activation region are of great significance for the regulation of anthocyanin biosynthesis genes, which helps to deepen the understanding of the regulation mechanism of anthocyanin biosynthesis pathway in cotton.Keywords: GhPAP1; transcription activation region; anthocyanin regulation; mutant.
