In this study, we aim to establish a cell model with conditional GR knockdown, and investigate the effect of icariin on GR expression. We plan to use CRISPR-Cas9 system to insert a specific small RNA into the GR gene, which leads to its degradation in cells and thus achieve the goal of conditional GR knockdown in cells. Meanwhile, different concentrations of icariin will be tested for their inhibitory effect on GR expression using Western blot and real-time fluorescence quantitative PCR techniques. The results may provide a better understanding of the biological function of GR and suggest new targets for disease treatment.Keywords: Glucocorticoid receptor (GR), conditional knockdown, cell model, icariin, inhibitory effect
