One of the pathological features of AD is the accumulation of amyloid-β (Aβ) peptides in brain tissue. Recent studies suggest that vaccination against Aβ may be a potential therapy for AD. However, previous clinical trials using full-length Aβ as the antigen have not shown satisfactory therapeutic benefits. To address this issue, we propose to develop a multi-epitope recombinant Aβ vaccine that targets multiple Aβ epitopes to elicit a more effective immune response.Objectives:1. To clone and express multiple epitopes of Aβ as recombinant fusion proteins2. To purify and assemble the recombinant fusion proteins into a multi-epitope vaccine3. To evaluate the immunogenicity of the multi-epitope vaccine in vitro and in vivoMethods:1. Bioinformatics analysis of Aβ epitopes to select candidate epitopes for the vaccine2. Coding genes for the selected epitopes will be synthesized and cloned into expression vectors3. Expression and purification of the epitope-fusion recombinant proteins4. Purification and assembly of the recombinant proteins into a multimeric vaccine5. Our study may offer a new approach for developing an effective therapeutic strategy for AD.Conclusion:The establishment of a multi-epitope recombinant Aβ vaccine and the evaluation of its immunogenicity will provide insight into the development of a more efficacious vaccine for AD.
