摘 要老化皮肤产生过量的活性氧自由基,本研究通过孵育过氧化氢(H2O2)建立小鼠成纤维细胞 L929 皮肤衰老模型,以羊栖菜多酚提取物为研究对象,实验羊栖菜多酚对 H2O2 诱导 L929 细胞氧化损伤的保护作用。结果表明;L929 细胞暴露于H2O2后,细胞活力会下降至参照组的 41.4%。随添加羊栖菜多酚浓度的升高,L929细胞活力逐渐上升,说明羊栖菜多酚对 H2O2诱导的 L929 细胞氧化损伤具有一定的保护作用。同时研究发现羊栖菜多酚可剂量依赖性降低 L929 细胞内活性氧自由基含量,提高 L929 细胞中总抗氧化能力,并可降低细胞内丙二醛含量,缓解细胞氧化损伤。本研究为羊栖菜多酚在抗衰海洋化妆品中的应用提供了理论支撑。关键词:羊栖菜多酚;小鼠成纤维细胞;过氧化氢;氧化损伤Abstract In this study, the mouse fibroblast L929 skin aging model was established by incubation with hydrogen peroxide (H2O2), and the protective effect of Sargassum fusiforme polyphenols on oxidative damage of L929 cells induced by H2O2 was studied. The results showed that Cell viability dropped to 41.4% of the control group after exposure to H2O2 . With the increase of the concentration of Sargassum fusiforme polyphenols, the activity of L929 cells increased gradually, which indicated that Sargassum fusiforme polyphenols had a protective effect on the oxidative damage of L929 cells induced by H2O2. Meanwhile, it was found that polyphenols from hijiki can reduce the content of ROS in L929 cells in a dose-dependent manner.increase the total antioxidant capacity in L929 cells, and reduce the content of malondialdehyde in cells to relieve oxidative damage to cells.This study provides theoretical support for the application of Sargassum fusiforme polyphenols in anti-aging marine cosmetics.
