相思树悬浮细胞培育及其细胞形态学观察摘要试验分析了基因型、愈伤组织类型、2,4-D 浓度、蔗糖浓度,起始接种量等各因素对相思树悬浮细胞培育的影响。结果表明:选择淡黄色,质地鲜嫩松脆的愈伤组织作为相思树悬浮培育的材料,能较快地建立起悬浮细胞系。黑木相思细胞小,胚性强,分散性好,最容易建立起悬浮细胞系。当蔗糖浓度为 30g/L,2,4-D 浓度为 0.5mg/L 时,适宜于悬浮细胞的保持。起始接种量对相思悬浮细胞系生长也有很大影响,从保持悬浮细胞系的角度看,每瓶 25mL 培育基中适宜的接种量为 0.5~1.0g。台湾相思悬浮细胞培育周期以 7d 继代 1 次,卷荚相思悬浮细胞培育周期以 8~10d 为宜。同时,通过对悬浮细胞培育的显微观察可以看出不同基因型的相思树悬浮细胞,其细胞生长状态各不相同。关键词相思树;悬浮细胞;细胞学观察中图分类号 S687;Q813.1 文献标识码 AAbstractTheeffectsofdifferentgenotypes,callustypes,concentrationsof2,4-D,sucroseconcentrations,andinoculationamountonthecellsuspensionculturewereanalyzedintheexperiment.TheresultsshowedthatthebestinitialmaterialoftheAcaciaspp.forcellsuspensionculturewasthefreshyellow,looseandsoftcallusIIandⅢ,whichwerepropitioustoestablishthesuspensioncellculturesystemquickly.Inaddition,thecellsoftheA.melanoxylonweresmall,ofstrongembryogeniccharacteristicsandfinedispersion,whichwereeasytoestablishthecellsuspensionsystem.Thecellsuspensioncouldbemaintainedwellundertheconditionsthatthesucroseconcentrationwas30g/Landthe2,4-Dconcentrationwas0.5mg/L.Andtheinitialinoculationamountalsoinfluencedthecultureofcellsuspension,ofwhichthebestinoculationamountwas0.5-1.0gin25mLcallieachbottle.Thesuitablesubculturecycleofthecellsuspensionculturewas7dinA.confusa,while8-10dinA.concinnata.Bythehistologicalobservation,itshowedthatthereweredifferentcellgrowthindifferentgenotypes.KeywordsAcaciaspp.;cellsuspension;histologicalobservationDOI10.3969/j.issn.1000-2561.2024.09.016相思树是中国引种的重要速生树种,在中国南方短周期工业原料林进展、水土保持和丰富林木种质资源等方面具有重要作用和地位[1]。植物细胞悬浮培育是植物细胞生长的微生物化,由于其分散性好,细胞形状及细胞...
