捷安肽素抗真菌作用机理讨论【摘要】 目的 讨论捷安肽素的抗真菌作用机理。方法 采纳形态学方法和同位素标记法。显微形态观察经捷安肽素处理后的供试真菌的形态学变化。进一步采纳 14C 同位素标记的特异底物“尿苷二磷酸--葡萄糖”示踪,讨论捷安肽素对真菌(1,3)-β-D-葡聚糖合成酶活性反应的影响。结果 显微形态观察表明,经捷安肽素处理后,供试真菌呈现球形膨胀,而且随着捷安肽素作用强度增强,球形膨胀的程度和发生频率也增加,提示捷安肽素的作用位点在真菌细胞壁。同位素标记实验表明,捷安肽素可抑制-β-D-葡聚糖合成酶的活性,降低真菌细胞壁合成过程中-β-D-葡聚糖的放射性掺入,对疫霉菌和辣椒炭疽病菌-β-D-葡聚糖合成酶的抑制率分别达到%和%。结论 捷安肽素的抗真菌作用机理为:抑制细胞-β-D-葡聚糖合成酶的活性,破坏真菌细胞壁的合成,造成真菌细胞壁组分-β-D-葡聚糖的缺损和病原真菌生长抑制。【关键词】 捷安肽素;抗真菌作用机理;-β-D-葡聚糖合成酶 A Research on Mechanism of Antifungal Polypeptide Jiean-peptide DAI Fu-ying1,ZHOU Jin-yan2,TAN Hong2 Abstract:Objective To investigate the antifungal mechanism of The mode of action of Jiean-peptide against 4 phytopathogenic filamentous fungi associated with phytopathy:Phytophthora sp.,Colletotrichum gossypii,Southw Botrytis cinerea and Fusarium oxysporum was investigated by the method of morphology and isotope (1,3)-β-D-glucan synthase assay was processed using isotope labelled substrate—urdine diphosphate(14C)glucose to study the effect of Jiean-peptide on fungal(1,3)-β-D-glucan Micrographic observation revealed that the Jiean-peptide made the hyphal expanded and (1,3)-β-D-glucan synthase assay showed Jiean-peptide could inhibit the activity of(1,3)-β-D-glucan synthase and reduce the radioactivity incorporation of(1,3)-β- inhibition percentages of Jiean-peptide against Phytophthora Colletotrichum gossypii Southw were % and % The antifungal mechanism of Jiean-peptide was the inhibition of enzyme activity of(1,3)-β-D-glucan synthase....
