题名页地震后危重多发伤伴混合感染病例分析张山,李思,王文武微生物与感染编辑部,上海200032基金项目:国家高新技术研究发展计划(863)(2009ZZ06…)第一作者:张山,详细联系方式(电话、E-mail)通信作者:王文武,详细联系方式(电话、E-mail)地震后危重多发伤伴混合感染病例分析张山,李思,王文武微生物与感染编辑部,上海200032摘要:为证实乙型肝炎病毒(hepatitisBvirus,HBV)3022nt-1787nt缺失突变体编码蛋白TS'X'(源于DNA聚合酶读码框架,T代表TP区,S'为部分缺失的spacer区,X'为截短的X蛋白)具有抗干扰素(IFN-)作用并确定其功能区域,首先以聚合酶链反应(PCR)扩增获得TS'X'全长及片段TS'(含TP区及部分缺失的spacer区),并克隆于pcDNA3.1/HisC载体。重组载体以FuGENE6转染Huh7肝细胞,48h后裂解细胞,通过融合表达的多肽表位抗体,以蛋白质印迹法(Westernblot)证实目的蛋白可在Huh7肝细胞中表达。重组载体及空载体pcDNA3.1/HisC分别与IFN-反应报告载体p6-16CAT按分子数5:1、10:1、15:1、30:1共转染Huh7肝细胞,转染后48h给予终浓度为100IU/ml的IFN-2a刺激,作用24h后裂解细胞,酶联免疫吸附试验(ELISA)检测胞内氯霉素乙酰基转移酶(CAT)含量。结果显示,与空白载体相比,随着TS'X'及TS'重组表达载体转染量的递增,Huh7胞内CAT值逐渐降低(n=6,P<0.05),但TS'X'与TS'之间无显著差异(n=6,P>0.05)。本研究证实HBV3022nt-1787nt缺失突变体编码的TS'X'蛋白可抑制Huh7细胞对IFN-的反应性,其活性与其N端TP及部分缺失的Spacer区有关。关键词:海分枝杆菌;转座子突变;斑马鱼;肉芽肿───────────────基金项目:国家高新技术研究发展计划(863)(2009ZZ06…)通信作者:王文武Correspondingautor.WANGWen-Wu,E-mail:jmi@fudan.edu.cnCasesanalysisofseveremultipletraumawithmixedinfectionsamongearthquakevictimsZHANGShan,LISi,WANGWen-WuJournalofMicrobesandInfection,Shanghai200032,ChinaAbstract:Thepurposeofthecurrentstudyistoinvestigatetheanti-IFN-effectsandtodeterminethefunctionalregionofthenovelproteinTS'X'encodedbythe3022nt-1787ntdeletionmutantofhepatitisBvirus(HBV).RegionscodingforTS'X'(inframewiththeopeningreadingframeofDNApolymerase,inwhichTstandsforterminalprotein,S'forpartiallydeletedspacerregion,andX'fortruncatedXprotein)andTS'(N-terminalofTS'X'containingterminalproteinandpartiallydeletedspacerregion)wereamplifiedbymeansofPCRandclonedintothepcDNA3.1/HisCvectorseparately.TherecombinantvectorwastransfectedintoHuh7hepatocytesindividuallybyFuGENE6transfectionreagent,andtheexpressionofthefusionproteinwasverifiedbyWesternblotanalysis.Therecombinantoremptyvectorwasco-transfectedtoHuh7hepatocyteswithIFN-responsereporterplasmidp6-16CATatthemolarratioof5:1,10:1,15:1and30:1,cellsweretreatedwithIFN-2a48hourspost-transfection.After24hoursofstimulation,thecellswerelysedandtheintracellularCATvaluewascalculatedbyELISAassay.Theresultsdemonstratedthat,ascomparedtotheemptyvector,theintracellularCATvaluesweregraduallyreducedasparalleledwithanincreasingamountofTS'X'orTS'recombinant(n=6,p<0.05),whilenosignificantdifferenceswereobservedbetweenthem(n=6,p<0.05).ItwasconcludedthatthenovelproteinTS'X'encodedbythe3022nt-1787ntdeletionmutantofHBVsuppressedtheresponseofHuh7hepatocytestoIFN-,andtheN-terminalregionwasafunctionaldomainforitsanti-IFN-effects.Keywords:Mycobacteriummarinum;Transposon;Zebrafish;Granuloma长久以来,分枝杆菌的细胞壁被认为是其致病性中的重要部分,他不仅可以帮助结核菌抵抗多种恶劣的理化及宿主体内环境,有利于结核菌抵抗大多数常用的抗生素,而且已证实多种细胞壁成分与其毒力密切相关[1,2]。此外,多种的抗结核药物,如异烟肼和乙胺丁醇,作用的靶位点...
