中性粒细胞淋球菌感染模型的建立来源:医学论文发表——赛恩斯作者:陈嵘,陈 蕾,张国学,樊翌明,吴志华作者单位: (广东医学院附属医院皮肤科,广东湛江【摘要】 目的 研究诱导型一氧化氮合酶(iNOS)及一氧化氮 (NO)在中性粒细胞淋球菌感染模型中的表达。方法分离正常人外周血中性粒细胞,在 10%胎牛血清 DMEM 培养液上培养3 h,A 组加入空白培养基, B 组加入淋球菌菌液,随后在0、3、8、12 h 分别以实时定量荧光 PCR 测定 iNOS mRNA 表达,用镉还原法检测NO 浓度。结果 B 组中 iNOS mRNA 表达和 NO 水平均较 A 组升高 (P<0.01),NO 浓度在 8 h 达到高峰。结论 中性粒细胞淋球菌感染模型可成功模拟该菌体内微氧感染环境,可用于研究淋球菌致病机制。【关键词】淋球菌 ;中性粒细胞 ;一氧化氮Establishment of gonococcusinduced neutrophil infection model CHEN Rongyi, ZHANG Guoxue, CHEN Lei, FAN Yiming, WU Zhihua (Department of Dermatology, Affiliated Hospital of Guangdong Medical College, Zhanjiang 524001, China) Abstract:Objective To investigate the expression of inducible nitric oxide synthase (iNOS) and nitric oxide (NO) in gonococcusinduced neutrophil infection model. Methods Peripheral blood neutrophils were islolated from health men, and then cultured in DMEM medium containing 10% fetal calf serum for 3 h. Blank medium or gonococcal inocula was added into group A or group B. NO concentration and iNOS mRNA expression at 0, 3, 8 and 12 h were determined by cadmium reduction method and realtime quantitative fluorescent PCR, respectively.Results Compared with group A, iNOS mRNA expression and NO level increased obviously in group B (P<0.01), and NO content peaked at 8 h.Conclusion The gonococcus induced neutrophil infection model can simulate the microoxygen infection circumstance in vivo, and is suitable for studying the pathogenic mechanism of gonococcus.
