The expression of AdipoR1 is regulated by various transcription factors, including Krüppel-like factor 15 (KLF15), which is highly expressed in adipose tissue and skeletal muscles. Previous studies have shown that KLF15 regulates AdipoR1 expression, but the precise regulatory mechanisms remain unclear.Objective: The aim of this study is to investigate the regulatory mechanisms of KLF15 on AdipoR1 expression using transcriptome analysis.Method: We will collect adipose tissue and skeletal muscle samples from wild-type mice and KLF15 knockout mice. Total RNA will be extracted, and RNA sequencing will be performed to analyze the transcriptome profiles of the samples. Differential expression analysis will be conducted to identify genes that are differentially expressed between the two groups. Functional enrichment analysis will be performed to explore the biological pathways that are associated with the differentially expressed genes. The expression levels of candidate genes will be validated by qPCR.Expected Results: We expect to identify genes that are differentially expressed between wild-type mice and KLF15 knockout mice. Functional enrichment analysis will reveal the biological pathways that are associated with the differentially expressed genes. Furthermore, our study will provide insights into the regulatory mechanisms of KLF15 on AdipoR1 expression.Conclusion: Our study will provide a better understanding of KLF15-regulated AdipoR1 expression and will contribute to the development of new therapeutic strategies for metabolic disorders such as obesity and type 2 diabetes.
