精品文档---下载后可任意编辑Peroxynitrite 诱导人脑血管平滑肌细胞 DDR2 表达机制的讨论的开题报告摘要:本讨论旨在探讨过氧亚硝酸(peroxynitrite,ONOO-)对人脑血管平滑肌细胞(HBVSMC)中 Discoidin Domain Receptor 2(DDR2)的表达及其机制的影响。通过实验模拟缺氧缺血和氧化应激的损伤情况,模拟器内注入 ONOO-,同时检测 HBVSMC 中 DDR2 的表达情况和相关的信号通路蛋白表达情况如 JNK/c-jun,p38 MAPK 和 NF-κB。结果显示,ONOO-可以显著增加 HBVSMC 中 DDR2 的表达,并激活 JNK/c-jun,p38 MAPK 和 NF-κB 通路,这些结果表明 ONOO-通过活化多条信号通路,诱导 DDR2 基因的表达,进而影响 HBVSMC 的发生,并对脑血管收缩和扩张可能产生影响。关键词:过氧亚硝酸; Discoidin Domain Receptor 2;血管平滑肌细胞;信号通路Abstract:The aim of this study is to investigate the effect of peroxynitrite (ONOO-) on the expression of Discoidin Domain Receptor 2 (DDR2) and its mechanism in human brain vascular smooth muscle cells (HBVSMC). The experimental conditions of hypoxia-ischemia and oxidative stress were simulated, and ONOO- was injected into the simulator. The expression levels of DDR2 and related signaling pathway proteins such as JNK/c-jun, p38 MAPK and NF-κB were detected in HBVSMC. The results showed that ONOO- significantly increased the expression of DDR2 in HBVSMC and activated JNK/c-jun, p38 MAPK and NF-κB pathways. These results suggest that ONOO- induces the expression of DDR2 gene by activating multiple signaling pathways, which may affect the occurrence of HBVSMC and the contraction and dilation of brain blood vessels.Keywords: peroxynitrite; Discoidin Domain Receptor 2; vascular smooth muscle cells; signaling pathway
