This study aims to analyze the characteristics of the integration site of exogenous genes mediated by φC31 integrase in the bovine genome and its potential applications. First, EGFP was inserted into the bovine genome using φC31 integrase, and two integration sites were selected for analysis. The results showed that the insertion site was stable and had no obvious effect on gene expression. Secondly, the expression level of φC31 integrase and the screening conditions of high-temperature heat were optimized. It was found that the expression level of φC31 integrase was the highest at 42℃ and 6 hours of cultivation. Finally, a transgenic bovine line with a functional gene was constructed by stable integration of exogenous genes mediated by φC31 integrase in the bovine genome. This study provides a theoretical and practical basis for the stable integration of exogenous genes in the bovine genome mediated by φC31 integrase.Keywords: φC31 integrase, transgenic, integration site, stable integration.
