摘 要谷胱甘肽过氧化物酶(GSH-Px)是机体内重要的氧化酶类之一。大黄鱼是我国重要的海水经济鱼类,也是宁德市的特色产业之一。本实验通过在大黄鱼饲料中添加不同比例的中草药复方(太子参 巴戟 当归 枸杞 茯苓 白术、、、、、),饲喂 30天后,以脾脏组织为实验材料,采用实时荧光定量 PCR 技术分析饲喂中草药后GSH-Px 基因的相对表达变化。结果显示:与对照组相比,各实验组中该基因的表达均出现了不同程度的下降(E5 组除外),E3 组的表达量下降最多(43%),其次是 E1 组(31%),E5 组的表达量比对照组升高 19%,但是各组间均无显著性差异(P 大于 0.05)。本研究结果为中草药作为饵料添加剂在大黄鱼饲料中的应用提供了基础数据,也为中草药作为鱼类免疫增强剂提供了前期研究基础。关键词:大黄鱼;基因表达;GSH-PxAbstractGlutathione peroxidase (GSH-Px) is one of the most important peroxidase enzymes in the body, which has an important effect on the innate immunity of fish. Larimichthys crocea is one of the most important economic fish in China and sea water is also one of the characteristic industry in Ningde. In this study, adding different proportion of Chinese herbal medicine compound were added in the feed of L. crocea (radix pseudostellariae, ba ji, angelica, medlar, poria cocos, atractylodes and so on). After feeding after 30 days, the spleen was isolated and extracted for RNA. The real-time fluorescent quantitative PCR was used to analyze the relative expression of GSH in different groups. The results showed that compared with the control group, the gene expression in each experiment group were decreased (with the exception of E5 group), and the expression of E3 group decreased most (43%), followed by E1 group (31%), the expression of E5 group amount higher than the control group was 19%, but there was no significant difference between groups (P is greater than 0.05).
