抗CD28抗体协同刺激增强抗CD3抗体体外激活T淋巴细胞并降低TGF

抗 CD28 抗体协同刺激增强抗 CD3 抗体体外激活 T 淋巴细胞并降低 TGF【摘要】 为了探讨体外激活 T 淋巴细胞的方法及活化后 T 淋巴细胞的免疫学特征，利用抗 CD3、抗 CD28 抗体在体外刺激健康人外周血单个核细胞 PBMNC，检测淋巴细胞转化，CD8+CD25+细胞比例和肿瘤生长因子 β 表达等相关免疫学指标。结果表明，在抗 CD28 抗体的协同作用下，抗 CD3 抗体的刺激活性明显提高，CD8+CD25+细胞比例明显增加，TGF-β 的表达下调。结论： 抗CD28 抗体协同刺激可以提供第二信号，使T 淋巴细胞充分活化，TGF-β 表达水平明显下降【关键词】 抗 CD3 抗体 Anti-CD28 Antibody Costimulation Enhances Anti-CD3 antibod Activating T Cells and Lowering TGF-β Expression in Vitro Abstract In order to study how to activate T cells and their immunological characteristics，the anti-CD3 and anti-CD28 McAbs were used to stimulate PBMNC，then their related immunological changes，such as lymphocyte transformation function，the percentage of CD8+CD25+ cells and TGF-β expression were delected lymplocyte transformation assay，flow cytometry and RT- PCR respectively. The results showed that in costimulation with anti-CD28 antibody stimulathetion，the activity of anti-CD3 antibody was significantly enchanced，the ratio of CD8+CD25+ cells of T cells was obviously increared，while TGF-β expression was down-regulated. It was concluded that the anti-CD28 antibody costimulation could provide stimulatory signal Ⅱ，which make T cells more active，while the expression of TGF-β significantly down-regulated.
