三种新冠病毒核酸提取方法的效果评价【摘要】目的通过比较三种核酸提取方法,了解不同新冠病毒核酸提取方法的性能。方法采用核酸释放剂法、磁珠法和离心吸附柱法对 10 份浓度为 500copies/ml的新冠病毒阳性质控品和 58 例新冠病毒核酸检测双阳性的鼻咽拭子样本进行核酸提取,提取后的核酸在同一品牌的荧光定量 PCR 扩增仪上,使用同一品牌的新冠病毒核酸扩增试剂进行 RT-PCR 扩增,对三种新冠病毒核酸提取方法的效果进行比较。结果试验结果显示,离心吸附柱法核酸提取效率最高,磁珠法次之,核酸释放剂法最低。结论离心吸附柱法提取核酸的效果最好,但操作繁琐、耗时,适合对阳性结果的复核。其次是磁珠法,既可以满足临床要求,又可以实现自动提取,适合于大样本的检测。核酸释放法虽然节省了核酸提取步骤和操作时间,但在三种方法中核酸提取的效率最低。关键词:新型冠状病毒;核酸提取;实时荧光定量 PCREvaluationofthreemethodsforextractingCOVID-19nucleicacidLIANGQihe,LIANGWanzhen,LIUJianting(ClinicallaboratoryofJiangmenCentralHospital,Jiangmen529070,China)[abstract]ObjectivetoinvestigatetheperformanceofdifferentCOVID-19nucleicacidextractionmethodsbycomparingthreenucleicacidsextractionmethods.Methodsnucleicacidsfrom10positiveCOVID-19QCsampleswith500copies/mlconcentrationand58positivecoronavirusnasopharyngealswabsampleswereextractedbynucleicreleaser,magneticbeadmethodandspincolumnmethod.TheextractednucleicacidswereamplifiedbyRT-PCRusingthesameCOVID-19nucleicacidamplificationreagentonthesamefluorescentquantitativePCRanalyzer,TheeffectsofthreeCOVID-19nucleicacidextractionmethodswerecompared.ResultsThetestresultsshowedthatthespincolumnmethodhadthehighestextractionefficiency,followedbymagneticbeadmethod,andtheextractionefficiencyofnucleicreleaserwasthelowest.ConclusionThespincolumnmethodhasthebestperformancefornucleicacidextraction,butitistediousandtime-consumingandsuitableforthereexaminationofpositivepatients.Thesecondisthemagneticbeadmethod,whichcanbothmeettheclinicalrequirementsandrealizeautomaticextraction,suitableforthedetectionoflargesamples.Nucleicreleaseralthoughsavingnucleicacidextractionstepsandoperatingtim...
