The biosynthesis of anthocyanins is regulated by a complex of three transcription factors, MYB, bHLH, and WD40, which are known as the MBW complex. The MBW complex regulates the expression of structural genes involved in the anthocyanin biosynthesis pathway. However, the mechanism of transcriptional regulation of anthocyanin biosynthesis in Yangmei remains poorly understood.Objectives:The main objective of this study is to investigate the transcriptional regulation mechanism of anthocyanin biosynthesis in Yangmei by the MYB-bHLH-WD40 complex. Specifically, the study aims to:- Identify MYB, bHLH, and WD40 genes involved in the anthocyanin biosynthesis pathway in Yangmei.- Determine the expression patterns of MYB, bHLH, and WD40 genes during fruit development and under different environmental conditions.- Analyze the interactions among MYB, bHLH, and WD40 proteins in regulating anthocyanin biosynthesis.- Investigate the effect of overexpression or knockdown of MYB, bHLH, or WD40 genes on anthocyanin accumulation in Yangmei.Methodology:- RNA sequencing (RNA-Seq) will be used to identify MYB, bHLH, and WD40 genes in Yangmei and analyze their expression patterns.精品文档---下载后可任意编辑- Protein-protein interaction assays such as yeast two-hybrid and bimolecular fluorescence complementation (BiFC) will be used to investigate the interactions among MYB, bHLH, and WD40 proteins in regulating anthocyanin biosynthesis.- Overexpression or knockdown assays using RNA interference (RNAi) or CRISPR-Cas9 technology will be performed to investigate the effect of MYB, bHLH, or WD40 genes on anthocyanin accumulation in Yangmei.- Anthocyanin content will be measured using high-performance liquid chromatography (HPLC) analysis.Expected outcomes:This study is expected to provide new insights into the transcriptional regulation mechanism of anthocyanin biosynthesis in Yangmei by the MYB-bHLH-WD40 complex. The identification of key genes and their expression patterns during fruit development and under different environmental conditions will enhance our understanding of the regulation of anthocyanin biosynthesis. The results of this study could be useful in developing strategies for enhancing anthocyanin accumulation in Yangmei and other plants.
