The bone marrow is a primary tissue affected by alcohol consumption, as it is responsible for hematopoiesis and can give rise to mesenchymal stem cells (MSCs). MSCs have the ability to differentiate into various cell types, including osteoblasts, which are responsible for bone formation. The impact of ethanol on the functions of MSCs and osteoblasts is still not fully understood.Objective:This study aims to investigate the effects of ethanol on the expression of neuropeptides and apoptosis-related genes in human bone marrow mesenchymal stem cells.Methods:Human bone marrow mesenchymal stem cells will be isolated and cultured in vitro. The cells will be treated with different concentrations of ethanol for different time periods to mimic chronic alcohol exposure. The expression of neuropeptides, including substance P, calcitonin gene-related peptide (CGRP), and tyrosine hydroxylase (TH), will be measured using quantitative polymerase chain reaction (qPCR) and enzyme-linked immunosorbent assay (ELISA). Apoptosis-related genes, including Bcl-2, Bax, and caspase-3, will also be measured using qPCR.Expected Outcomes:It is expected that ethanol exposure will lead to changes in the expression of neuropeptides and apoptosis-related genes in human bone marrow mesenchymal stem cells. These findings may have implications for the development of new therapies to prevent or treat alcohol-induced bone loss and fractures.
