Recent studies have shown that CNA interacts with proteasome subunit PSMA6, which has an important role in the function of proteasomes. However, the specific mechanism of this interaction is still unclear. Therefore, this study aims to systematically explore the mechanism of interaction between CNA and PSMA6.Multiple techniques will be used to investigate the interaction between CNA and PSMA6, including protein-protein interaction, mass spectrometry, structural biology and cellular biology techniques. Firstly, we will use GST pulldown and CoIP techniques to verify the interaction between CNA and PSMA6. Secondly, we will determine the binding site between them by mass spectrometry. Then, we will resolve the interaction mode between them by X-ray crystallography. Finally, we will conduct cellular biology studies to investigate the effect of CNA on the function of PSMA6.It is expected that this study will clarify the interaction mechanism between CNA and PSMA6 and its effect on the function of proteasomes, providing an important theoretical and practical basis for further understanding the biological function of proteasomes and the pathogenesis of related diseases.Keywords: Calcineurin A, Proteasome subunit PSMA6, Interaction mechanism, Protein-protein interaction, Mass spectrometry, Structural biology, Cellular biology.
