Although several molecular mechanisms have been proposed for NPC initiation and progression, the exact etiology is still unclear. The identification of candidate tumor suppressor genes and oncogenes is critical for understanding the pathogenesis of NPC and developing potential therapeutic targets.NPCEDRG (Nasopharyngeal carcinoma-associated expressed sequence 1 down-regulated gene) is a novel candidate tumor suppressor gene discovered in our previous study. Our preliminary data showed that NPCEDRG was frequently downregulated in NPC tissues and cell lines, and its low expression was associated with poor prognosis of NPC patients. However, the underlying mechanism of NPCEDRG regulation in NPC remains largely unknown.Objectives:The aim of this study is to investigate the regulatory mechanism of NPCEDRG expression in NPC cells and explore its role in NPC progression.Methods:1. NPC cell culture: NPC cell lines (CNE1, CNE2, 5-8F, HNE1) will be cultured in RPMI-1640 medium supplemented with 10% fetal bovine serum.2. Transfection and RNA interference: NPC cells will be transfected with pcDNA-NPCEDRG plasmid or siRNA targeting NPCEDRG using lipofectamine 3000 reagent. The efficiency of transfection will be confirmed by qRT-PCR and Western blot.3. qRT-PCR: The mRNA expression levels of NPCEDRG and related genes (such as c-Myc, p53, and E2F1) will be measured 精品文档---下载后可任意编辑by qRT-PCR. The relative mRNA levels will be calculated using the 2^-ΔΔCt method.4. Western blot: The protein expression levels of NPCEDRG and its downstream targets will be detected by Western blot analysis. GAPDH will be used as an internal control.Expected results:We expect to identify the critical regulators of NPCEDRG expression in NPC cells and evaluate their effect on NPCEDRG-mediated tumor suppression. Our data may shed light on the understanding of the molecular mechanisms underlying NPC occurrence and progression, and provide new insights into the diagnosis and treatment of NPC.
