抗丙型肝炎病毒第一高变区抗体捕获 HCV 病毒的实验讨论【摘要】 目的 采纳抗丙型肝炎病毒第一高变区抗体，建立病毒捕获实验。方法 重组表达 HVR1 嵌合抗原，免疫家兔，制备抗 HVR1 抗体，与 20 例患者阳性血清反应，通过 HCV RT-PCR 检测捕获结果。结果 在 30 例样品中有 13 例完全捕获，16 例部分捕获 ，仅 1 例未捕获。结论 抗 HVR1 多抗可高交叉捕获 HCV 病毒。【关键词】 肝炎病毒丙型； 互补决定区抗体； 聚合酶链反应 Studies on the antibodies against hypervariable 1(HVR1) of hepatitis C virus (HCV) capture virus 【Abstract】 Objective A set of test capture virus was formulated. Using antibodies against hypervariable 1(HVR1) of hepatitis C virus (HCV). Methods Recombinant expression antigens of chimeric HVR1, the preparation of antibodies against HVR1 by immuno-rabbit,with 30 cases patients of HCV infection capture test，the results were detected by HCV RT-PCR. Results In 30 cases of HCV infection can capture virus of 13cases completely, 16 cases capture partly,only 1case can not capture. Conclusions The antibodies against hypervariable 1(HVR1) of hepatitis C virus (HCV) can broadly capture virus of HCV.【Key words】 hepatitis C virus; complementarity determining regions antibody; polymerase chain reaction 丙型肝炎是由丙型肝炎病毒感染引起的世界性传染病。讨论表明，约 70％～85%的 HCV 感染将进展为慢性肝炎，其中部分最终将进展成为肝硬化和肝细胞癌，是极为严重的世界公共卫生问题。而目前仍没有有效的治疗和预防的方法，问题在于 HCV变异大，而中和性表位所在的 HVR1 区变异更大，变异引起的免疫逃逸，要获得有效的免疫几乎不可能。但最近讨论表明，HVR1 虽然高变，但仍有一定规律性，存在着一定的免疫交叉反应性，又为这方面讨论带来一线曙光。于是就出现了有关高交叉反应性 HVR1 的讨论，也取得了可喜的结果。Zibert［1］等证明 HVR1-Ab 能有效捕获同株病毒粒子，Zhou［2］等证明 HVR1-Ab 能阻止同株病毒吸附于培育细胞上。本讨论用克隆表达的交叉性 HVR1 作为免疫原，制备抗 HVR1 抗...
