Intercellular adhesion molecule-1 (ICAM-1) and lymphocyte function-associated antigen-1 (LFA-1) are key molecules involved in immune cell adhesion and activation, and their expression levels may affect the outcome of islet cell transplantation.Objective: The objective of this study is to investigate the relationship between ICAM-1/LFA-1 expression levels and the outcome of islet cell transplantation, in order to provide a theoretical basis for the improvement of transplantation efficacy.Methods: Islet cells will be isolated and purified from Sprague-Dawley rats, and randomly divided into experimental and control groups. The experimental group will be treated with inflammatory factors to induce ICAM-1/LFA-1 expression, while the control group will be treated with saline. The expressions of ICAM-1/LFA-1 will be detected by flow cytometry, and the adhesion of immune cells to islet cells will be evaluated by adhesion assays. The islet cells of both groups will also be transplanted into diabetic rats, and the blood glucose levels and survival rates of the rats will be observed for 4 weeks.Expected Results: We expect that the expression levels of ICAM-1/LFA-1 will be higher in the experimental group than in the control group, and that immune cell adhesion to islet cells will be stronger in the former. We also expect that the blood glucose levels and survival rates of diabetic rats receiving islet cell transplantation from the experimental group will be lower than those receiving transplantation from the control group.Conclusion: This study aims to explore the relationship between ICAM-1/LFA-1 expression and the outcome of islet cell transplantation, in order to provide a basis for the improvement of transplantation efficacy.
