精品文档---下载后可任意编辑SiRNA 沉默 XIAP 基因对骨肉瘤 MG-63 细胞生物学特性的影响中期报告摘要目的:探讨 SiRNA 沉默 XIAP 基因对骨肉瘤 MG-63 细胞生物学特性的影响。方法:选取人类骨肉瘤 MG-63 细胞作为讨论对象,通过随机分组实验,分别对无处理组、阴性对比组、脂质体对比组和 SiRNA 处理组进行处理,采纳 RT-PCR 和 Western blotting 方法检测 XiapmRNA 和 Xiap蛋白表达水平。采纳 MTT 法检测各处理组的细胞增殖情况,采纳Annexin V-FITC/PI 双染尝法检测细胞凋亡率。最后,采纳细胞迁移实验和基质侵袭实验检测细胞迁移和侵袭能力。结果:经过 SiRNA 干预治疗,MG-63 细胞中的 XiapmRNA 和Xiap 蛋白表达均得到抑制,同时细胞增殖率随着 SiRNA 干预治疗时间的延长而减少,细胞凋亡率逐渐增加。此外,SiRNA 处理组的细胞迁移和侵袭能力明显下降。结论:SiRNA 沉默 XIAP 基因可以抑制骨肉瘤 MG-63 细胞增殖,促进细胞凋亡,并且降低细胞迁移和侵袭能力。关键词:SiRNA;XIAP 基因;骨肉瘤;生物学特性AbstractObjective: To investigate the effect of siRNA-mediated silencing of XIAP gene on the biological characteristics of osteosarcoma MG-63 cells.Methods: Human osteosarcoma MG-63 cells were selected as the research object. Randomized grouping experiment was conducted to treat no treatment group, negative control group, liposome control group and SiRNA treatment group. RT-PCR and Western blotting method were used to detect Xiap mRNA and Xiap protein expression levels. MTT assay was used to detect cell proliferation in each treatment group, and Annexin V-FITC/PI double-staining method was used to detect cell apoptosis rate. Finally, cell migration and invasion ability were detected by cell migration and matrix invasion experiments.Results: After SiRNA intervention, the expression of Xiap mRNA and Xiap protein in MG-63 cells was inhibited, and the 精品文档---下载后可任意编辑cell proliferation rate decreased with the prolongation of SiRNA intervention time, and the cell apoptosis rate gradually increased. Moreover, the migration and invasion ability of SiRNA treatment group were significantly reduced.Conclusion: SiRNA-mediated silencing of XIAP gene can inhibit the proliferation of osteosarcoma MG-63 cells, promote cell apoptosis, and reduce cell migration and invasion ability.Keywords: SiRNA; XIAP gene; osteosarcoma; biological characteristics.
