The aim of this study was to establish a rapid method for measuring serum 1,5-AG and to apply it to clinical detection. UPLC-MS technology was used for analysis, and the pretreatment, separation, and detection parameters were optimized to establish a rapid, accurate, and reproducible method. After validation, the method had a good linear range (0.01-100 μg/mL), detection limit (0.01 μg/mL), correlation coefficient (r>0.999), accuracy (97.30%-102.80%), and precision (RSD<3.28%). The method was applied to analyze the serum 1,5-AG concentrations of 60 healthy individuals and 60 diabetes patients. It was found that the 1,5-AG concentration in diabetes patients was significantly lower than that in healthy individuals (P<0.05) and was related to glycemic control.Keywords: 1,5-anhydroglucitol; UPLC-MS; new biochemical marker; early screening for diabetes.
