The aim of this study is to construct a human CEA gene vaccine and validate its expression in tongue cancer cell lines. The human CEA gene was amplified by PCR, cloned into the pUC19 cloning vector, and confirmed for correctness by restriction enzyme digestion and sequencing. The pVAX1-CEA recombinant plasmid was constructed by cloning the human CEA gene into the expression vector pVAX1. The recombinant plasmid was transfected into the tongue cancer cell line SCC-15 using the latex method, and its expression in the transfected cell line was detected by Western blot. The results of the experiment showed that the human CEA gene was successfully cloned into the expression vector and successfully transfected into the tongue cancer cell line, which expressed the target protein.Keywords: human CEA gene; vaccine; tongue cancer cell line; expression
