?实验研究Ciliaryneurotrophicfactor-loadedpolyethyleneglycol-polylactic-co-glycolicacidmicrobubblescombinedwithultrasoundforprotectionofretinalganglioncellsWANGWen-ting1,LIUSu1*,WANGZhi-gang2,ZHOUXi-yuan1(1.DepartmentofOphthalmology,theSecondAffiliatedHospitalofChongqingMedicalUniversity,Chongqing400010,China;2.InstituteofUltrasonicImaging,ChongqingMedicalUniversity,Chongqing400010,China)[Abstract]ObjectiveToprepareandobservethepropertiesofciliaryneurotrophicfactor-loadedpolyethyleneglycol-polylactic-co-glycolicacidultrasoundcontrastagents(CNTF-PEG-PLGA),andtoassesstheprotectioneffectofCNTF-loadedPEG-PLGAmicrobubblescombinedwithultrasoundonretinalganglioncells(RGCs)afteropticnerveinjuryinrats.MethodsCNTF-loadedPEG-PLGAultrasoundmicrobubbleswerepreparedusingdoubleemulsiontechnique,andthepropertiesoftheultrasoundmicrobubbleswereobserved.Totally145adultSDratswererandomlydividedinto7groups.Afteropticnerveinjury,RGCswereretrogradelabeledwiththefluorescenttracerfluorogold(FG)tocountRGCsnumber.Theexpressionsofgrowthassociatedprotein43(GAP-43)weredetectedbyimmunohistochemistryonthe3rdday,the1st,the2ndandthe4thweekafteropticnerveinjury.Theretinalpathologicalmorphologychangewasobservedatdiffer-enttimes.ResultsCNTF-loadedPEG-PLGAultrasoundmicrobubblesweresmoothandsphericalwithameansizeof(312.5±57.35)nm,withthedrugloading0.298μg/mgandencapsulationefficiencyof62.35%.Therewere93.60%CNTF-PEG-PLGAreleasewithin28days.TheresultsshowedthatafterRGCswereretrogradelabeledwithFG,theRGCscountofgroupGwashigherthanthatofothergroupsateverytime,(allP<0.05),butlessthangroupA(P<0.05).TheexpressionofgroupGwashigherthanthatofothergroups(P<0.05)onthe1st,the2ndandthe4thweekaf...
