精品文档---下载后可任意编辑Aβ 对星形胶质细胞增殖与活化作用的体外讨论的开题报告摘要阿尔茨海默病(AD)是一种老年性疾病,其主要病理特征是脑内 β 淀粉样蛋白(Aβ)的异常沉积。星形胶质细胞是中枢神经系统的重要组成部分,其在 Aβ 致 AD发生进展中起着重要的作用。本讨论旨在探究 Aβ 对人脑星形胶质细胞增殖和活化的影响。采纳人脑星形胶质细胞株 U251,观察不同浓度 Aβ(0、5、10、20、40 μmol/L)对其增殖和活化的影响。细胞增殖率采纳 MTT 法检测,细胞活化情况采纳ELISA 法测定白细胞介素 1β 和肿瘤坏死因子-α 的含量。估计结果是,Aβ 会抑制星形胶质细胞的增殖,但同时也会促进其活化,导致白细胞介素 1β 和肿瘤坏死因子-α 的分泌量增加。该讨论对了解 Aβ 致 AD 的机制及其对星形胶质细胞的影响有重要意义。关键词:阿尔茨海默病;星形胶质细胞;β 淀粉样蛋白;增殖;活化AbstractAlzheimer's disease (AD) is an aging disease characterized by the abnormal deposition of β-amyloid protein (Aβ) in the brain. Astrocytes are an important part of the central nervous system and play an important role in the pathogenesis and development of AD induced by Aβ. This study aims to investigate the effect of Aβ on the proliferation and activation of human astrocytes.U251 human astrocyte cell line was used to observe the effect of different concentrations of Aβ (0, 5, 10, 20, 40 μmol/L) on their proliferation and activation. The cell proliferation rate was detected by MTT method, and the cell activation was determined by ELISA method for the content of interleukin-1β and tumor necrosis factor-α.It is expected that Aβ will inhibit the proliferation of astrocytes, but also promote their activation, leading to an increase in the secretion of interleukin-1β and tumor necrosis factor-α. This study is of important significance for understanding the mechanism of Aβ induced AD and its effect on astrocytes.Keywords: Alzheimer's disease; astrocytes; β-amyloid protein; proliferation; activation
