精品文档---下载后可任意编辑ISG15 蛋白稳定表达细胞系的构建及其与猪源BVDV--2 相互作用初步讨论的开题报告【摘要】ISG15 蛋白是一种干扰素-stimulated 基因(ISG)产物,可以调节免疫反应和细胞增殖,并在许多病毒感染和细胞外压力中发挥重要作用。本讨论旨在构建 ISG15 稳定表达细胞系,并初步讨论其与猪源 BVDV-2的相互作用。通过经经过筛选后的侧翼肌细胞(ISG15 +/+),构建 ISG15 稳定表达细胞系,并采纳 Western blot 鉴定 ISG15 的表达水平。同时,利用ELISA 和 Real-time PCR 进行猪源 BVDV-2 的检测和分析,探究 ISG15蛋白与猪源 BVDV-2 的相互作用特点。通过本讨论的开展可为进一步探究 ISG15 蛋白的功能及其在病毒感染发生过程中的分子机制提供新思路和参考。【关键词】ISG15; 稳定表达; 猪源 BVDV-2; 相互作用【Abstract】ISG15 protein is a product of interferon-stimulated genes (ISGs), which can regulate immune responses and cell proliferation, and play important roles in many virus infections and extracellular stresses. The aim of this study is to construct an ISG15 stable expression cell line and to preliminarily study its interactions with porcine BVDV-2.ISG15 stable expression cell lines were constructed by side muscle cells (ISG15+/+) that had been screened. The expression level of ISG15 was identified by Western blot. Meanwhile, ELISA and Real-time PCR were used for detection and analysis of porcine BVDV-2 to explore the characteristics of interactions between ISG15 protein and porcine BVDV-2.By carrying out this study, new ideas and references are provided for further exploration of the function of ISG15 protein and its molecular mechanism in the process of virus infection.【Keywords】ISG15; stable expression; porcine BVDV-2; interaction
