Thus, cloning, expression and activity analysis of the D15D gene is important on better understandings the metabolism pathway of HUFA in organisms.Objectives:The objectives of this study is to clone, express, and analyze the activity of the D15D gene from marine invertebrates, in order to explore its potential roles in HUFA synthesis.Methods:1. Cloning of D15D gene:Total RNA will be isolated from the tissue samples of marine invertebrates using Trizol reagent. The complementary DNA (cDNA) will be synthesized by reverse transcription of RNA using MMLV reverse transcriptase. The D15D gene will be obtained by PCR amplification using gene-specific primers. The PCR products will be purified and then cloned into expression vectors, such as pET-28a or pcDNA3.1 (+), to construct recombinant plasmids.2. Expression of D15D gene:The recombinant plasmids will be transformed into expression hosts, such as E. coli or CHO cells, and then induced to express the D15D protein. The expressed protein will be purified using affinity chromatography.3. Activity analysis of D15D protein:The activity of D15D will be determined by measuring the conversion of precursor fatty acids to HUFA using gas chromatography or liquid chromatography-mass spectrometry 精品文档---下载后可任意编辑(LC-MS) analysis. The optimum enzyme conditions, such as temperature and pH, will be determined to maximize D15D activity. The effects of different substrates and inhibitors on the activity of the D15D will be investigated.Expected results:We expect to successfully clone, express and purify the D15D protein from marine invertebrates. The optimized conditions for enzyme activity will be established, and the kinetics of the enzyme will be explored, which will potentially lead to improved production of HUFA in aquaculture industry.Conclusion:The investigation of the D15D gene cloning, expression and activity analysis, will provide a better understanding of the metabolism pathway in marine organisms, and bring interesting implications to the development of aquaculture industry.
