The UL31 protein encoded by the HSV-1 genome is an important structural protein playing key roles in virus infection and replication, and nuclear aggregates formation of the host cell. However, the role of host proteins interacting with UL31 in the virus life cycle is unclear.This study aims to identify the host protein interacting with HSV-1 UL31 and investigate its biological functions. By protein affinity purification and mass spectrometry analysis, we have identified the host protein TMEM140 interacting with UL31, which is involved in HSV-1 entry and replication. Furthermore, we will use cell culture media and primary cells to evaluate the role of TMEM140 in the HSV-1 lifecycle and to reveal its biological functions. The results of this research may contribute to understanding the mechanism of HSV-1 infection.Keywords: HSV-1, UL31, TMEM140, interaction, biological function
