精品文档---下载后可任意编辑丹参多酚酸盐干预肝纤维化大鼠糖代谢异常的实验讨论的开题报告摘要:本实验旨在探究丹参多酚酸盐对肝纤维化大鼠糖代谢异常的干预作用及其机制。40 只雄性 SD 大鼠随机分为 4 组:正常组、模型组、丹参多酚酸盐低剂量干预组和高剂量干预组。采纳 CCl4 诱导肝纤维化模型,对各组大鼠进行体重监测、OGTT、ITTT、OGIS 测试、胰岛素序贯检查、病理组织学观察及相关分子对比实验。结果显示,与模型组相比,丹参多酚酸盐低剂量和高剂量干预组大鼠的 OGTT、ITTT、OGIS、血浆胰岛素、HOMA-IR 等指标均有明显改善,其中高剂量组效果较佳。病理组织学观察显示,丹参多酚酸盐可以明显减轻肝纤维化程度。分子对比实验结果显示,丹参多酚酸盐干预组的糖代谢相关基因(GLUT2, GLP-1, SGLT1, G6pc)的表达明显高于模型组。因此,丹参多酚酸盐能够通过影响糖代谢相关基因的表达来改善肝纤维化大鼠的糖代谢异常,从而具有潜在的治疗和预防肝纤维化的作用。关键词:丹参多酚酸盐,肝纤维化,糖代谢异常,机制讨论。Abstract:This experiment aims to explore the intervention effect and mechanism of Danshen polyphenolic acid on glucose metabolism disorder in rats with hepatic fibrosis. Forty male SD rats were randomly divided into four groups: normal group, model group, low-dose intervention group and high-dose intervention group of Danshen polyphenolic acid. CCl4 was used to induce hepatic fibrosis model. The weight monitoring, OGTT, ITTT, OGIS test, insulin sequential examination, pathological histological observation, and molecular control experiment were performed on each group of rats. The results showed that compared with the model group, OGTT, ITTT, OGIS, plasma insulin, HOMA-IR and other indicators of rats in low-dose and high-dose intervention groups of Danshen polyphenolic acid were significantly improved, and the effect of high-dose group was better. The pathological histological observation showed that Danshen polyphenolic acid could significantly reduce the degree of hepatic fibrosis. The results of molecular control experiment showed that the expression of glucose metabolism related genes (GLUT2, GLP-1, SGLT1, G6PC) in Danshen polyphenolic acid intervention group was 精品文档---下载后可任意编辑significantly higher than that in model group.
