精品文档---下载后可任意编辑侵染大蒜 12 种病毒 CP 基因的原核表达、抗血清制备及其病毒检测的开题报告【摘要】大蒜是我国的重要蔬菜作物之一,然而其生长过程中易遭受病毒的侵染,导致产量和品质受到影响。本讨论旨在利用原核表达技术和抗血清制备技术,制备 12 种侵染大蒜的病毒 CP 基因的抗血清,并应用于病毒检测,为大蒜的病毒预防和控制提供技术支持。首先根据先前文献报道,分别从侵染大蒜的病毒中克隆出相应的 CP 基因序列,然后利用 PCR技术构建重组质粒,并将其转化至大肠杆菌中进行原核表达。接着采纳SDS-PAGE 和 Western blot 检测表达的蛋白质,并经过免疫小鼠制备抗血清。最后应用 ELISA 和免疫电镜检测病毒感染的大蒜。估计通过本讨论,可以制备出多种侵染大蒜的病毒 CP 基因的抗血清,并建立一系列检测方法,为大蒜病毒的防治开拓新途径。【关键词】大蒜;病毒;CP 基因;原核表达;抗血清;病毒检测【Abstract】Garlic is an important vegetable crop in China, but it is easily infected by viruses during its growth, resulting in reduced yield and quality. This study aims to use prokaryotic expression technology and antiserum preparation technology to prepare antiserums against the CP genes of 12 viruses that infect garlic, and apply them to virus detection, providing technical support for virus prevention and control of garlic. First, the corresponding CP gene sequences were cloned from the viruses that infect garlic according to previous literature. Then, the recombinant plasmid was constructed using PCR technology and transformed into Escherichia coli for prokaryotic expression. The expressed proteins were detected by SDS-PAGE and Western blot, and antiserums were prepared by immunizing mice. Finally, the garlic infected by viruses was detected by ELISA and immunoelectron microscopy. It is expected that this study will prepare antiserums against the CP genes of multiple viruses that infect garlic and establish a series of detection methods, thereby developing new ways for the prevention and control of garlic viruses.【Keywords】Garlic; Virus; CP gene; Prokaryotic expression; Antiserum; Virus detection
