精品文档---下载后可任意编辑食管肿瘤干细胞分离及生物学性状的初步讨论的开题报告【摘要】食管肿瘤干细胞的讨论对于食管癌的治疗和预防具有重要意义。本讨论旨在通过分离和鉴定食管肿瘤干细胞,探究其生物学性状与肿瘤发生、进展的机制。首先采纳流式细胞仪(FACS)将颗粒细胞分离出CD133 阳性的细胞群,并通过限制性稀释法得到单克隆细胞系。接着利用免疫荧光染色和 Western Blot 分析 CD133 阳性细胞的特异性表达,观察其增殖、分化、侵袭能力及肿瘤形成能力,并分析其在促肿瘤因子作用下途径及信号通路的变化。本讨论对于食管肿瘤干细胞的深化认识和相关治疗策略的探究具有一定的理论和实际意义。【关键词】食管肿瘤干细胞;CD133;生物学性状;肿瘤形成【Abstract】The study of esophageal tumor stem cells is of great significance for the prevention and treatment of esophageal cancer. The aim of this study is to isolate and identify esophageal tumor stem cells, and to explore their biological characteristics and the mechanism of tumor occurrence and development. Firstly, the granular cells were separated from CD133 positive cell population with flow cytometry (FACS), and then single clone cell lines were obtained by limiting dilution method. Then, the specific expression of CD133 positive cells was analyzed by immunofluorescence staining and Western Blot, and their proliferation, differentiation, invasion ability and tumor formation ability were observed. The changes in pathway and signaling pathway under the action of tumor-promoting factors were analyzed. This study has certain theoretical and practical significance for further understanding of esophageal tumor stem cells and the exploration of related treatment strategies.【Keywords】 esophageal tumor stem cells; CD133; biological characteristics; tumor formation
