精品文档---下载后可任意编辑RGD 接枝氧化海藻酸钠 N-琥珀酰壳聚糖复合水凝胶的制备及体外诱导分化讨论的开题报告摘要:随着干细胞技术的快速进展,干细胞治疗已成为一个备受关注的讨论热点。而干细胞的体外培育中,细胞外基质(ECM)具有重要的作用,应用 ECM 复合材料来促进干细胞分化进展成为一种重要的方法。本讨论旨在制备一种 RGD 接枝氧化海藻酸钠 N-琥珀酰壳聚糖复合水凝胶,并对其进行体外诱导分化讨论。首先通过紫外光谱、红外光谱和核磁共振等方法验证所制备水凝胶的化学结构,并通过扫描电子显微镜观察其形貌;其次进行降解实验,测试水凝胶在酶体内的降解情况;最后进行细胞实验,比较水凝胶与普通壳聚糖水凝胶对人类成骨细胞(hBMSCs)分化的影响,讨论水凝胶对 hBMSCs 分化的诱导作用。关键词:RGD 接枝;水凝胶;琥珀酰壳聚糖;体外诱导分化Abstract:With the rapid development of stem cell technology, stem cell therapy has become a research hotspot. In the in vitro culture of stem cells, extracellular matrix (ECM) plays an important role. The application of ECM composite materials to promote stem cell differentiation has become an important method. The aim of this study is to prepare an RGD-grafted oxidized sodium alginate N-succinyl chitosan composite hydrogel and study its in vitro induced differentiation. Firstly, the chemical structure of the prepared hydrogel was verified by UV spectroscopy, infrared spectroscopy and nuclear magnetic resonance, and its morphology was observed by scanning electron microscopy. Secondly, degradation experiments were carried out to test the degradation of the hydrogel in lysosomes. Finally, cell experiments were carried out to compare the effects of the hydrogel and ordinary chitosan hydrogel on the differentiation of human bone marrow mesenchymal stem cells (hBMSCs), and to study the induction effect of the hydrogel on hBMSCs differentiation.Keywords: RGD grafting; hydrogel; succinyl chitosan; in vitro induced differentiation
