PCB 降糖片的纯化工艺及体外药效研究内容摘要:研究大孔树脂精制 PCB 醇提液中总黄酮,水提液中多酚、多糖的工艺条件。方法:把 PCB 的总黄酮吸附率和解析率作为指标,研究 4 种型号完全不同的大孔树脂对 PCB 总黄酮的吸附力及与解析力,确认 D101 作为最适树脂。优化 D101 大孔树脂的纯化条件;将 PCB 多酚、多糖吸附率与解析率作为指标,研究 5 种不同类型大孔树脂对 PCB 多酚、多糖的吸附力和解析力,将 AB-8 作为最适树脂。优化 AB-8 大孔树脂纯化条件。结果:PCB 总黄酮分离富集条件为:上样速度 2 BV/h,上样量 120.00 mL,上样浓度为 4.890 mg/mL,吸附 1.5 h,除杂溶剂 4%乙醇,除杂速度 3 BV/h,用量 256.41 mL,将 60%乙醇作为洗脱溶剂,3 BV/h 的洗脱速度,125.00 mL 洗脱剂用量,经 D101 处理后 PCB 总黄酮纯度至55. 25% ;多酚、多糖的分离富集条件为:上样速度 2 BV/h,上样量 15.00 mL,上样浓度为 0.5 g/mL(生药/溶剂),1.5 h 吸附,40%乙醇作为洗脱溶剂,3 BV/h 的洗脱速度,90.00 mL 洗脱剂用量,经 AB-8 处理后的 PCB 多酚、多糖纯度为 10.76%、40.92%,结论:D101 型的大孔吸附树脂能较好将 PCB 总黄酮纯化富集;AB-8 型大孔树脂能较好将 PCB 多酚、多糖纯化富集。关键词:PCB 降糖 纯化工艺Study on purification technology and in vitro efficacy of PCB Jiangtang tabletsAbstract: the research of macroresin PCB ethanol extract of total flavonoids in liquid water extraction of polyphenols, process conditions of polysaccharides.
