下载后可任意编辑两株肿瘤细胞系 HLA I 类分子基因表达水平的鉴定及意义【摘要】 目的 探讨两株常见肿瘤细胞系 Hela 细胞和 BEL 7402细胞 HLA I 类分子基因表达水平及其 影响 CTL 反应性的可能意义。 方法 提取肿瘤细胞的基因组DNA,利用 PCR 反应在基因组水平鉴定HLA I 类分子基因的拷贝数情况;提取细胞总 RNA, RT PCR反应合成 cDNA,利用 PCR反应鉴定肿瘤细胞 HLA I 类分子在 RNA 水平的表达情况。结果 两株肿瘤细胞的 HLA I类基因在基因组水平的拷贝数无明显下降;下载后可任意编辑在 RNA 水平上,Hela 细胞的 HLA A 、HLA B和 HLA C 基因均有表达,表达水平无明显下降,BEL 7402细胞的 HLA A 和 HLA C 基因有表达,但 HLA C 基因表达水平降低,而HLA B 基因未见明显表达。 结论 RNA 水平上,部分 HLA I 类分子基因表达的降低或缺失推测可能是导致 BEL 7402细胞比 Hela 细胞出现 CTL 反应性下降的原因之一。 【关键词】 HLA I 类分子;Hela 细胞;BEL 7402细胞;mRNA Abstract:Objective To investigate 下载后可任意编辑the expression level of HLA I molecule genes in two tumor cell lines, Hela and BEL 7402 and the possible significance of CTL Genome DNA were extracted from these two tumor lines for detecting the HLA I molecule genes′ levels by RNA from two cell lines were extracted for RT PCR, HLA I molecule genes′expression on RNA level were judged by PCR and agarose gel HLA I molecule genes levels did 下载后可任意编辑not decrease significantly on genome RNA level, HLA A, HLA B and HLA C all expressed normally without significant descent in Hela cell line.
