In this study, we aim to isolate, purify, and clone the chitinase gene from Bacillus sp. S65.Objectives:1. Isolation and identification of Bacillus sp. S65 from environmental samples.2. Optimization of culture conditions for chitinase production.3. Purification of chitinase from the culture supernatant using chromatography techniques.4. Cloning and sequencing of the chitinase gene from Bacillus sp. S65.5. Expression of the cloned chitinase gene in a heterologous host.Methodology:1. Isolation and identification of Bacillus sp. S65:a. Environmental samples (soil, water, and sediment) will be collected from different locations and cultured on nutrient agar.b. Bacterial colonies will be screened for chitinase activity using colloidal chitin as a substrate.c.
