The use of laser capture microdissection (LCM) to purify NPC tissues enables the specific analysis of tumor cells without contamination from surrounding normal tissues. In this study, we aim to establish protein expression profiles for LCM-purified NPC tissues and normal nasopharyngeal epithelial tissues in order to identify differentially expressed proteins and potential biomarkers for NPC.(Introduction)Nasopharyngeal carcinoma is a malignant tumor arising from the epithelial cells lining the nasopharynx. It has a high incidence rate in Southern China, Southeast Asia, and North Africa. Despite advances in radiotherapy and chemotherapy, the prognosis of NPC remains poor. Therefore, the identification of reliable biomarkers for the early detection and prognosis of NPC is of great importance. However, current diagnostic markers for NPC lack sensitivity and specificity, and there is a need for the identification of novel biomarkers for improving the diagnosis and treatment of NPC.LCM is a technique that enables the isolation of specific cells or tissues from complex samples. In the case of NPC, LCM can be used to specifically capture tumor cells from biopsy samples, enabling the analysis of protein expression profiles of NPC tissues without contamination from surrounding normal tissues. By comparing the protein expression profiles of LCM-purified NPC tissues and normal nasopharyngeal epithelial tissues, differentially expressed proteins can be identified as potential biomarkers for NPC.(Methods)In this study, we will use LCM to isolate NPC tissues and normal nasopharyngeal epithelial tissues from biopsy samples. The purity of the isolated tissues will be assessed by H&E staining and microscopic examination. The isolated tissues will be subjected to protein extraction and 精品文档---下载后可任意编辑trypsin digestion. The resulting peptides will be analyzed by liquid chromatography tandem mass spectrometry (LC-MS/MS) for protein identification and quantification. The protein expression profiles of LCM-purified NPC tissues and normal nasopharyngeal epithelial tissues will be compared using bioinformatics tools to identify differentially expressed proteins.(Expected Results)We expect to establish protein expression profiles for LCM-purified NPC tissues and normal nasopharyngeal epithelial tissues. We will identify differentially expressed proteins between the two tissue types, which may serve as potential biomarkers for NPC. The identification of reliable biomarkers for NPC will improve the diagnosis and treatment of this disease.
