In this paper, we propose a new method, namely the remote thiyl-assisted NCL reaction, which can be used to achieve chemical synthesis and modification of proteins.This method first introduces an auxiliary thiol-protecting group at the C-terminus of the protein, and then uses the NCL reaction to remove the thiol-protecting group from the protein while connecting the target molecule with the protein.
