This study aims to explore the cloning and copy number of Mx1 gene in different ploidy of common carp, laying a foundation for further research on its functions. Mx1 gene fragments of 2n, 3n and 4n carp were isolated and analyzed by PCR amplification and sequencing, and sequence alignments and phylogenetic tree construction were carried out. Meanwhile, the copy numbers of Mx1 genes in different ploidy carp were determined by qPCR. The results showed that the Mx1 gene sequence lengths of different ploidy carp were similar, their evolutionary distances were close, and they basically belonged to the same subtype, with 3n carp having the highest diversity. The qPCR results showed that the copy numbers of Mx1 gene were higher in 3n carp, followed by 4n carp, and lowest in 2n carp. These results provide a theoretical basis and experimental basis for further studying the functions and antiviral mechanisms of Mx1 gene in carp.【Key words】Mx1 gene; common carp; ploidy; cloning; copy number
