摘 要用秋水仙素低渗空气干燥法,制备福建福鼎市虎纹蛙骨髓细胞染色体玻片,并利用 Giemsa 染色对 Howell 和 Black 的快速银染法稍加改进。研究福建福鼎市 地区虎纹蛙染色体组型和 Ag-NORs,结果表明:其核型公式为 2n=26,NF=52,可配成 13 对染色体组;其中大型染色体 5 对(No.1~5), 小型染色体 8 对(No.6~13),未发现次缢痕和与性别相关的异型性染色体,在 Ag-NORs 中未观察到银染点。通过与已报道不同地区的虎纹蛙的染色体组型和 Ag-NORs 相关资料比较,结果表明不同地区虎纹蛙的染色体亚中部着丝点(SM)、染色体数目和次缢痕、随体的位置等方面都存在着一定差异,说明其核型具有多样性。关键词:福建福鼎市;虎纹蛙;染色体组型;Ag-NORs;多样性AbstractThe colchicine hypotonic air drying method was used to prepare chromosome slides of bone marrow cells of tiger frogs in Fuding City, Fujian Province, and the rapid silver staining method of Howell and Black was slightly improved by Giemsa staining. Studying the karyotype and Ag-NORs of tiger frogs in Fuding City, Fujian Province, the results show that the karyotype formula is 2n = 26, NF = 52, which can be matched into 13 pairs of chromosomes; 5 of them are large chromosomes (No.1 ~ 5), 8 pairs of small chromosomes (No.6 ~ 13), no subconstriction and sex-related atypical chromosomes were found, and silver stains were not observed in Ag-NORs. Compared with the reported karyotypes and Ag-NORs data of tiger frogs in different regions, the results showed that the chromosomes of sub-central centromere (SM), chromosome number and subconstriction marks, satellites of tiger frogs in different regions There are certain differences in location and other aspects, indicating that their karyotypes are diverse.
