Recently, studies have shown that this enzyme is encoded by a small gene family in some species of plants. However, there is limited research on how APX small gene families in poplar, specifically the 741 Populus tomentosa APX (PtoAPX) gene, function.Objectives:The main aim of this study is to clone the PtoAPX small gene family and analyze their biochemical characteristics. The objectives include:1. Amplification and cloning of the PtoAPX small gene family2. Expression analysis of the PtoAPX small gene family in response to abiotic stress3. Biochemical characterization of the cloned PtoAPX small gene familyMethodology:1. RNA extraction and cDNA synthesis: RNA will be extracted from the leaves of Populus tomentosa using the RNeasy Plant Mini Kit, and cDNA will be synthesized using the RevertAid First Strand Synthesis Kit.2. Amplification and cloning of PtoAPX small gene family: Specific primers will be designed using the PtoAPX nucleotide sequence available in the NCBI database. PCR will be performed using cDNA as the template, and the amplified products will be cloned into a pMD-19T vector.3. Expression analysis: The expression patterns of the PtoAPX small gene family will be analyzed using quantitative real-time PCR (qPCR) in response to abiotic stress.4. Biochemical characterization: The biochemical characteristics of the cloned PtoAPX small gene family will be analyzed through determining enzyme activity, protein purification, substrate specificity, and enzyme kinetics.Expected outcomes:The successful cloning of the PtoAPX small gene family and their subsequent biochemical characterization would help elucidate the mechanisms of ROS scavenging and stress responses in Populus tomentosa.
