Hib polysaccharide has the ability to induce an immune response and has been widely used in the preparation of Hib vaccines. The aim of this study is to optimize the preparation process of Hib polysaccharide and to study its binding with D protein. In the process of preparing Hib polysaccharide, the most suitable culture conditions were found through the comparison of different cultivation conditions, and further optimization of polysaccharide extraction, purification, and separation was carried out. By using methods such as ELISA and surface plasmon resonance technology, the binding of Hib polysaccharide with D protein was studied to analyze its binding ability and binding mode. This study is expected to provide important references for the preparation of Hib vaccines and related research on immune response.Key words: Hib; polysaccharide; D protein; preparation; binding
