FHF is often accompanied by high morbidity and mortality, therefore, it is urgent to search for effective treatment strategies for FHF. PDTC (Pyrrolidine dithiocarbamate) is a selective inhibitor of NF-κB, which is involved in many inflammatory and apoptotic responses. However, the effects of PDTC on NF-κB expression and cell apoptosis in FHF are still largely unknown. Therefore, this study aims to investigate the effects of PDTC on NF-κB expression and cell apoptosis in liver tissue of FHF rats.Methods:FHF rats were established by D-galactosamine (D-GalN) and lipopolysaccharide (LPS) injection. The rats were randomly divided into three groups: FHF group, PDTC group (FHF rats treated with PDTC) and control group (normal rats). After treatment, hepatic function, histopathology, cell apoptosis and NF-κB expression in the liver tissue were detected.Results:Compared with the control group, FHF group showed significant liver function damage and histopathologic changes. However, after PDTC treatment, the liver function was significantly improved and the histopathologic changes were relieved. The level of apoptosis in liver tissue was significantly decreased in the PDTC group compared with the FHF group. Moreover, the expression of NF-κB in liver tissue was significantly decreased in the PDTC group compared with the FHF group.Conclusion:PDTC effectively improve liver function and histologic pathology in the FHF rats.
