In this study, we investigated the population variation and phylogeography of Chinese scaly-tailed gecko using molecular ecology approaches. By collecting field samples, extracting DNA, performing PCR amplification and sequencing, we obtained D-loop region sequences of mitochondrial DNA from 44 individuals of the gecko, and conducted phylogenetic and population genetics analyses in combination with other related sequences in the GenBank public database. Our results showed that the Chinese scaly-tailed gecko has significant phylogeographical structure and high genetic differentiation among populations in different geographic regions. Further analysis revealed that this genetic differentiation may be related to topographic barriers and climate differences between mountainous and plain regions. This study has important implications for understanding the population history and evolution process of the Chinese scaly-tailed gecko.Keywords: Chinese scaly-tailed gecko; population variation; phylogeography; molecular ecology
